    Relative quantification of CYP1A gene expression in whitefish (Coregonus lavaretus) exposed to benzo[a]pyrene

    The expression of CYP1A (cytochrome P4501A) can be induced by a number of aromatic compounds in teleost fishes. We developed a real-time PCR assay for measuring relative quantities (RQ) of CYP1A mRNA in whitefish (Coregonus lavaretus). To test for the usefulness of the assay we performed a treatment study, using benzo[a]pyrene (B[a]P) a model CYP1A inducer. Primers for the CYP1A gene were adapted from the literature, whereas those for [beta]-actin (endogenous control) were designed from a region that was found to be conserved among salmonid [beta]-actin genes. A group of hatchery raised whitefish, with an average body mass of 15 g and total length of 12 cm were given an intraperitoneal injection (10 mg/kg) of B[a]P in corn oil (2 mg B[a]P/ml corn oil) or corn oil alone (Control). After 48 h, whitefish liver, head kidney and brains were collected for mRNA isolation and analysis. In all three tissues sampled, CYP1A mRNA was affected by treatment with B[a]P. Head kidney tissue showed the greatest induction potential (RQ=11.00) from base levels (RQ=1.00), followed by liver (RQ=9.45), and brain (RQ=3.76). These results demonstrated that CYP1A was highly inducible by B[a]P in whitefish head kidney and liver, and to some extent, in brain tissue. The approach presented here has the advantage of providing rapid and accurate measures of CYP1A induction in various tissues of fish responding to PAH contaminant exposure

    Past strong experiences determine acute cardiovascular autonomic responses to acoustic stress

    Background: Stress is a major risk factor for cardiovascular (CV) disease. We hypothesized that past strong experiences might modulate acute CV autonomic responses to an unexpected acoustic stimulus. A i m: The study’s aim was to compare acute CV autonomic responses to acoustic stress between students with and without a past strong experience associated with the acoustic stimulus. Materials and Methods: Twenty five healthy young volunteers - medical and non-medical students - were included in the study. CV hemodynamic parameters, heart rate (HR), and blood pressure (BP) variability were assessed for 10 min at rest and for 10 min after two different acoustic stimuli: a standard sound signal and a specific sound signal used during a practical anatomy exam (so-called "pins"). Results: Both sounds stimulated the autonomic nervous system. The "pins" signal caused a stronger increase in HR in medical students (69 ± 10 vs. 73 ± 13 bpm, p = 0.004) when compared to non-medical students (69 ± 6 vs. 70 ± 10, p = 0.695). Rises in diastolic BP, observed 15 seconds after sound stressors, were more pronounced after the "pins" sound than after the standard sound signal only in medical students (3.1% and 1.4% vs. 3% and 4.4%), which was also reflected by low-frequency diastolic BP variability (medical students: 6.2 ± 1.6 vs. 4.1 ± 0.8 ms2, p = 0.04; non-medical students: 6.0 ± 4.3 vs. 4.1 ± 2.6 ms2, p = 0.06). Conclusions: The "pins" sound, which medical students remembered from their anatomy practical exam, provoked greater sympathetic activity in the medical student group than in their non-medical peers. As additives can be made using the post-consumer wood, generated during wood products processing, re-use of recycled waste materials in the production of biocomposites can be an environmentally friendly way to minimalize and/or utilize the amount of the solid waste. Also, bioactive materials, which possess small amounts of antimicrobial additives belong to a very attractive packaging industry solution. This paper presents a study into the biodegradation, under laboratory composting conditions, of the composites that consist of poly[(R)-3-hydroxybutyrate-co-4-hydroxybutyrate)] and wood flour as a polymer matrix and natural filler, respectively. Thermogravimetric analysis, differential scanning calorimetry and scanning electron microscopy were used to evaluate the degradation progress of the obtained composites with different amounts of wood flour. The degradation products were characterized by multistage electrospray ionization mass spectrometry. Also, preliminary tests of the antimicrobial activity of selected materials with the addition of nisin were performed. The obtained results suggest that the different amount of filler has a significant influence on the degradation profile.Published onlin

    Neutrophil-to-lymphocyte ratio, platelet-to-lymphocyte ratio, and systemic immune-inflammation index as clinical predictive and prognostic markers in patients with advanced pancreatic cancer receiving gemcitabine monotherapy

    Introduction. Pancreatic cancer is characterized by an increasing incidence and still poor prognosis despite the availability of various therapeutic options, currently including single- and multi-drug chemotherapy as well as molecularly targeted therapy. Therefore, appropriate qualification for particular therapies, based mainly on clinical and histological factors, is extremely important. Inflammatory status, associated with cancer development, justifies the search for prognostic markers related to the immune system, which could be additional factors facilitating selection of appropriate therapy. This study aimed at assessing the prognostic value of the neutrophil-to-lymphocyte ratio (NLR), plateletto- lymphocyte ratio (PLR), and systemic immune-inflammation index (SII) in patients with advanced pancreatic cancer undergoing gemcitabine monotherapy.  Material andmethods. A retrospective analysis of blood morphological parameters was performed in 167 patients with advanced pancreatic cancer treated with gemcitabine monotherapy in the first line in five oncology centers in Poland in the years 2017–2022. The NLR, PLR, and SII were calculated, and cut-off points between high and low values were defined. Clinical parameters and their distribution were assessed depending on the overall survival (OS) value equal to or greater than or less than median OS. The distribution of patients within OS intervals in relation to the categories of inflammatory markers was assessed.  Results. The median age of patients was 71 years, the majority were women (58%), with clinical stage IV (57%), and with dominant location of metastases in the liver (42.5%). The median NLR was 2.69 (range 0.5–36.65), PLR 146.54 (range 18.53–1118.57), and SII 784.75 (range 79.86–10622.67). The cut-off points were defined as 4.5625 for the NLR [125 patients (75.8%) with a value less than and 40 patients (24.3%) with a value equal to or greater], 150 for the PLR [87 (52.7%)/ 78 (47.3%)], and 897.619 for the SII [96 (58.2%)/69 (41.8%)]. Comparing the groups with OS longer than or equal to the median and OS shorter than the median, statistically significant differences were found in relation to body mass index (BMI) (p = 0.02), baseline stage (p < 0.001), and location of metastases (p < 0.001). There were statistically significantly more NLR and SII values below the cut-off points in patients with survival at least equal to median OS. Concerning the PLR, no statistically significant differences were found between groups determined by OS value.  Conclusions. We demonstrated the relationship between indicators calculated on the basis of blood count parameters and treatment results. It may indicate the predictive and prognostic importance of indices reflecting immune system status, which can be a valuable addition to the clinical criteria included in prognostic models.

    Systemic treatment of patients with advanced pancreatic cancer — is there still a place for gemcitabine in the first-line setting? Experience of Polish oncology centers

    Introduction. Despite some progress in the treatment of patients with pancreatic cancer, it is still a malignancy with a poor prognosis, which results from its rapid local growth with a tendency to infiltrate surrounding tissues and metastasize, and late diagnosis at the advanced stage. The use of multi-drug regimens and modern targeted therapies did not completely eliminate the use of gemcitabine in monotherapy, which is a therapeutic option mainly in patients with poor performance status, ineligible for more advanced therapies. This study aimed to evaluate the results of treatment with single-agent gemcitabine in everyday clinical practice in Poland and to attempt to identify the predictors of obtaining long-term responses resulting from this treatment.  Material and methods. A retrospective analysis of 167 patients with advanced pancreatic cancer treated with single-agent gemcitabine in five oncology centers in Poland in the years 2017–2022 was conducted. Gemcitabine was used as monotherapy at an initial dose of 1000 mg/m2 of body surface area (BSA) weekly, 7 times in an 8-week cycle, then 3 times in a 4-week cycle.  Results. Median overall survival (OS) in the entire group of patients was 6.1 months (range — 0.2–32.3 months), and median progression-free survival (PFS) was 4.2 months (range — 0.2–31.3 months). A group of 60 patients was identified as “long responders” (LR), with a response of at least 6 months and a group of 107 as “short responders” (SR). Median PFS in the LR group was 9.15 months (range — 6.0–31.3 months) and in the SR group, it was 3.2 months (range — 0.2–5.8 months). Median OS was 11.6 months (range — 5.9–30.8) and 3.8 months (range — 0.2–32.3 months), respectively. In multivariate analysis, the likelihood of achieving at least a 6-month response (LR) was assessed using a logistic regression model. The model takes into account four variables: the neutrophil/lymphocyte (NLR) ratio, liver metastases, sex, and Hb level. Conclusions. The obtained results confirm that gemcitabine monotherapy is still useful in the first-line treatment of patients with advanced and metastatic pancreatic adenocarcinoma. An appropriate selection of patients for this treatment may improve the results while maintaining lower toxicity compared to combined treatment.

    Prioritization of Candidate Genes in QTL Regions for Physiological and Biochemical Traits Underlying Drought Response in Barley (Hordeum vulgare L.)

    Drought is one of the most adverse abiotic factors limiting growth and productivity of crops. Among them is barley, ranked fourth cereal worldwide in terms of harvested acreage and production. Plants have evolved various mechanisms to cope with water deficit at different biological levels, but there is an enormous challenge to decipher genes responsible for particular complex phenotypic traits, in order to develop drought tolerant crops. This work presents a comprehensive approach for elucidation of molecular mechanisms of drought tolerance in barley at the seedling stage of development. The study includes mapping of QTLs for physiological and biochemical traits associated with drought tolerance on a high-density function map, projection of QTL confidence intervals on barley physical map, and the retrievement of positional candidate genes (CGs), followed by their prioritization based on Gene Ontology (GO) enrichment analysis. A total of 64 QTLs for 25 physiological and biochemical traits that describe plant water status, photosynthetic efficiency, osmoprotectant and hormone content, as well as antioxidant activity, were positioned on a consensus map, constructed using RIL populations developed from the crosses between European and Syrian genotypes. The map contained a total of 875 SNP, SSR and CGs, spanning 941.86 cM with resolution of 1.1 cM. For the first time, QTLs for ethylene, glucose, sucrose, maltose, raffinose, α-tocopherol, γ-tocotrienol content, and catalase activity, have been mapped in barley. Based on overlapping confidence intervals of QTLs, 11 hotspots were identified that enclosed more than 60% of mapped QTLs. Genetic and physical map integration allowed the identification of 1,101 positional CGs within the confidence intervals of drought response-specific QTLs. Prioritization resulted in the designation of 143 CGs, among them were genes encoding antioxidants, carboxylic acid biosynthesis enzymes, heat shock proteins, small auxin up-regulated RNAs, nitric oxide synthase, ATP sulfurylases, and proteins involved in regulation of flowering time. This global approach may be proposed for identification of new CGs that underlies QTLs responsible for complex traits

    Assessment of Operational Performance for an Integrated ‘Power to Synthetic Natural Gas’ System

    This article presents a power to SNG (synthetic natural gas) system that converts hydrogen into SNG via a methanation process. In our analysis, detailed models for all the elements of the system are built. We assume a direct connection between a wind farm and a hydrogen generator. For the purposes of our calculations, we also assume that the hydrogen generator is powered by the renewable source over a nine-hour period per day (between 21:00 and 06:00), and this corresponds to the off-peak period in energy demand. In addition, a hydrogen tank was introduced to maximize the operating time of the methanation reactor. The cooperation between the main components of the system were simulated using Matlab software. The primary aim of this paper is to assess the influence of various parameters on the operation of the proposed system, and to optimize its yearly operation via a consideration of the most important constraints. The analyses also examine different nominal power values of renewables from 8 to 12 MW and hydrogen generators from 3 to 6 MW. Implementing the proposed configuration, taking into account the direct connection of the hydrogen generator and the methanation reactor, showed that it had a positive effect on the dynamics and the operating times of the individual subsystems within the tested configuration
